BACKGROUND:	Aak1Antibody:AP2-associatedproteinkinase1(Aak1)isamemberoftheArk1/Prk1subfamilyofSer/Thrproteinkinasesthatarethoughttoregulateendocytosisbyphosphorylatingtheaccessoryendocyticcomponents.Aak1interactswithandphosphorylatesthemu2subunitoftheAP-2complex,whichpromotesbindingoftheAP-2totyrosinebased(Yxxphi)internalizationmotif-containingreceptorsandsubsequentreceptorendocytosis.AtleasttwoisoformsofAak1areknowntoexist;thelongerisoformcontainsanextendedcarboxy-terminusthatcontainsanadditionalclathrin-bindingdomain.OverexpressionofthislongisoformorAak1depletionbyRNAinterferenceimpairstransferrinrecyclingfromtheearly/sortingendosome,suggestingthatAak1functionsatmultiplestepsoftheendosomalpathwaybyregulatingtransferrininternalizationanditsrecyclingbacktotheplasmamembrane.
